HCT116 and SW480 cells were transfected with shUKLFGFP or OV-UKLFGFP to construct UKLF knockdown or overexpression cell lines, (B) the mRNA and (C–D) protein expression of SLC39A4 was measured. (E) Analysis of UKLF binding sites in the SLC39A4 promoter by JASPAR database. (F) HCT116 cells were co-transfected with SLC39A4 promoter luciferase reporter plasmids and UKLF overexpression plasmids (OV-UKLF), 48 h post-transfection, the relationship between UKLF and SLC39A4 was determined by a luciferase reporter assay. (G) The binding of UKLF to the promoter regions of SLC39A4 was determined by chromatin immunoprecipitation. HCT116 were co-transfected with OV-UKLFGFP and shSLC39A4GFP, 48 h post-transfection, the mRNA expression of (H) UKLF and (I) SLC39A4 was detected by qRT-PCR. (J) The viability was measured with the MTT assay. Values were mean ± SD. *p < 0.05, **p < 0.01. Cell experiments were performed at least three biological repetitions per group.
